Thing is, hair follicles, sebaceous glands, sweat glands, etc. CANNOT grow in scar tissue, right? So if this all grows back as they suggest it does, that sounds pretty scar-free. At least to the point where any amount of scar tissue wouldn't be noticeable? That's how it sounds to me anyway. Getting those appendages to return is the hallmark of healing without scarring.
I am so excited, like a lot of you, but are things that I am confused about!
1. Like you mentioned, why is this not blowing up? They claim to be able to regenerate full thickness wounds - the implications of this would be so huge and widespread..... Why isn't this everywhere? Why aren't more people talking about it?
2. Human studies? Where are they? All of their recent displays of confidence would suggest that it has shown to be effective in humans, yet, we have not heard any details of this. Why?...? I want them now!! Lol.
3. This is not really a concern for me, but after so many years of waiting and waiting for promising research to come to fruition and hating the FDA for seemingly stifling all progress, SkinTE seems to have just flown through. I love how quick they are moving. Does this also indicate unprecedented effectiveness?
I have been burned (no pun intended) by similar excitement many times, but this has me feeling pretty optimistic... Ahhhh, tell us more PolarityTE.

The regenerated skin might be unrecognizable to normal skin, but no one would know for sure until they talk about human trials. Although, on their website, they show pictures of the healing process. Idk if that's using a pig model or a human model.

That's right they are lost because the Myofibroblasts present in the injury become scar tissue. It's the process our body chose so that it can heal itself quickly. think of it as a built in survival mechanism.
PolarityTE has achieved full thickness regeneration in all 3 layers of the skin including all of its appendages as well. So yes it is incredibly close to scar free.

The news was just posted less than 24 hours ago. Give it a couple more weeks. It is usually the case that forums like this are the first to know the second that any new information is published. We are the ones who look everywhere for it in the first place after all. I'll give you an example:
If I had no scarring and flawless vibrant skin, but instead blurry and diminishing vision, do you think I'd seek knowledge and information about skin? Probably not since whatever I have done up to that point has allowed me to maintain great skin. I'd probably seek to attain better vision though. Regardless any breakthrough discovery that is made a reality will become known to the world in time.

Human trials are either finishing up or have just recently finished. Since they've announced confirmation of market entry of their product by the end of this year, there will definitely be results and pictures published for the world to see. Just sip on a glass of wine until then.

Yes and No
The reason they were able to fly past the FDA is because they were using autologous tissue and homlogous factors to achieve regeneration. Cells are extracted from the healthy skin of the patient and then re-introduced into the defected area after it has gone through polarization. No risk of rejection either since the product came from the patients themselves. It doesn't necessarily mean "unprecedented effectiveness" because you have other treatments that also used a patient's own cells, but turned out to be shams just like PRP and Acell for example that seem to be no better than snake oils. The FDA is more so like a safety detective. In this case though, they demonstrated an incredibly effective solution that allowed them to bypass FDA phase trials and provide a revolutionizing product to the entire skin market.
Polarity used the right methodology and thus was able to make it a reality.

How do you think it differs from ReCell/ACell? I am worried the result will be similar. Also, what do you think of the skin biopsy? I really don't want another scar from where they take the biopsy.

That's tough to figure. I would only be able to say they simply got the formula right. They also may be using a different platform than the others. They have a 3D platform technology that allows the extracted cells to be imprinted into and undergo what they call "polarization." The specifics of that is unknown and they probably won't reveal that to avoid competitors stealing their tech. What I do know is that they base it on establishing a community of cells to work together around the injured/defected area to regenerate skin.
Although a skin biopsy is required, it's only a small amount. They'll have to expand more on the biopsy which they will soon. I do believe you will probably be able to take the biopsy from anywhere in your body that has healthy skin. We'll just have to wait a little longer an see for ourselves.
I wonder what the peoples' skin looks like that had this done. It must be incredible to them, being the first people without regenerated skin like that. I want that feeling so badly.
Would they publish photos of the people's faces and skin when the trials are concluded? Or does that go against the FDA's privacy and regulations of the patient/participant's confidentiality or something?
GUYS!! SUNOGEL PUBLISHED A PAPER LAST WEEK!!

Can you please explain by what you mean by "propagating"
HERE ARE THE MAIN NOTES OF THE SUNOGEL PAPER.
"Full-thickness skin wounds commonly lead to nonfunctioning scarring in adults. More than 100 million patients develop scars from elective surgeries per annum in industrialized countries. Besides, a much larger population is badly in need of scar reduction therapies."
-Recent advances in skin biology of scarless wound healing and biomaterial science enable us to regenerate perfect skin than ever before.
-Given the pro-regenerative potential of immune modulating scaffolds for wound healing, we set forth to develop dextran-based bioabsorbable hydrogels that can modulate immune responses by promoting M2 macrophage phenotype, and further attenuating scar formation during cutaneous wound healing.
-We then tested it for acute skin injuries using both murine and porcine models. We demonstrated that our innovative DexIEME hydrogel promoted complete skin regeneration on both pre-existing scarred skin and deep full skin injuries by producing mature epithelial structure with hair follicles and glands.
-A multiple functional scaffold with intricate structure could be instructional, but the complexity may hold back its translation to clinics due to increased cost and challenge in manufacturability while escalating the production. While designing hydrogel precursors, other than improving biocompatibility and regulating M2 macrophage phenotype, we also try to simplify the formulation to ensure product reproducibility, whereby we reduce formulation steps by increasing the self-crosslinking capacity here. In our design (Figure 1A), the increase in the length of side chain allows us to crosslink the hydrogel with itself. Unlike Dex-AE that has a relatively short side chain, which barely form a hydrogel with itself due to steric hindrance effect, DexIEME can self-crosslink to form a hydrogel.
-These hydrogel characteristics allow the skin stem cells to make new scaffolds and self-assemble into complete skin structures. To further examine its regenerative potential, we then tested the hydrogel to regenerate new skins in both murine and porcine models.
-Scar reduction treatment could benefit hundreds of millions of people that are suffering from scar pain.
-The higher magnification images shows that DexIEME is able to regenerate complete skin with appendage from the scar site in mice.
-Pig skin is very similar to human skin and pig is thus commonly used as a preclinical model for wound healing. To further demonstrate its regenerative capacity, we tested the hydrogel in deep full skin injury. The skin cut was down to the adipose layer. Full skin of A1 inch (2.5 cm) in diameter was first removed, and skin regenerated after 10 weeks (Figure 6A). The HE images (Figure 6B) further revealed the wound healing progress and the hydrogel clearly regenerated new skin on the pig. The hydrogel treated wound contained more adipose tissue than nonhydrogel treated wounds (n = 4) (Figure 6C). In contrast, the nonhydrogel treated wound (Figure 6Biii) showed more fibrosis than the hydrogel treated wound (Figure 6Bvi). The nonhydrogel treated wound developed fibrotic collagen, while the hydrogel-treated wound developed full skin structure with normal basket-weave-like collagen. Furthermore, the hydrogel treated wound is more mature, and the new skin retains a reticulated epithelial layer (Figure 6Bii,v). Clearly, the newly regrown skin is fully regenerated compared with normal skin (Figure S5, Supporting Information). This result strongly indicates the hydrogel potential in clinical wound cares.
-We first demonstrated that DexIEME hydrogel promoted complete skin regeneration with hair regrowth on pre-existing scars, while preclinical studies further demonstrated that our DexIEME hydrogel regenerated full skin by producing new hair follicles, epithelial reticulation, and differentiated dermal during deep porcine wound healing.
-Though the scar was created by burn injury, it is arguably possible that DexIEME could treat pre-existing scars created by other injuries. To further evaluate its regenerative capacity, we then tested in a preclinical model. Here, DexIEME hydrogels were able to regenerate full skin on acute deep skin injuries.
-Plikus et al. recently reported that the conversion of myofibroblasts into adipose cells aided scarless wound healing.
-Overall, our approach to investigate immune-modulated hydrogel yielded the development of a pro-regenerative Dex- IEME hydrogel. This hydrogel has the capacity to lead to greater success in skin regeneration and the attenuation of scar formation during deep wound healing. With fine-tuned formulation, our pro-regenerative DexIEME hydrogel regenerated full skin structures with appendages on both pre-existing scars and acute wounds in murine and porcine models. DexIEME would be a great candidate for wound repair and skin regeneration. In addition to promoting skin regeneration, dextran hydrogels can also serve as a carrier for genes, cytokines, and to manipulate other stimuli to treat various compromised skin diseases. The success of these efforts will undoubtedly improve human health and thus have great values.

It clearly showed scar-free healing on pre-existing scar. Even if remove it partly.
There's a lot of research going into scarless healing. It's a great time to be optimistic because many of us have scars we want to see gone. It will also benefit burn victims or people who have been disfigured by some unlucky event.
We also live in a society where beauty standards are more important than ever. Young girls and boys have a big pressure on their shoulders to look their best. Acne sucks. Having these little huge red pus filled bastards all over your body destroys your self esteem at such a young age. The scars it leaves behind ruin your confidence as a young adult when trying to find your place in this world.
Plastic surgeons are in high demand. More and more people want to look their best. It's sad as a society that we have come to this but we all tend to follow the masses. I believe if scarless healing is achieved this will benefit plastic surgeons the most as more people will ask to go under the knife without any scars.
However, we need to be cautious about what's out there on the Internet. Many on here have pointed out how before some other physicians have claimed to have regenerated skin but years passed and they are forgotten.
I believe there are many diseases out there which a cure has been found but big pharma tends to cover it up. Let's face it, they are not suppose to cure but rather treat. There's more money to be made. This is why I believe with scars it will be the same thing. They will treat your scars. This is what is being done today nonetheless for some minor improvement. Therefore, the promises made by SkinTE should be taken with a bit of caution.
They made promises about stem cells a decade ago and suddenly they are not mentioned that often. Let's face it, they still haven't found a cure for male pattern baldness. How some of you here believe scarless healing will be achieved with SkinTE is beyond me. Our bodies whenever we get injured always form scars. It's just how nature is. Wounds have to heal fast. Well if you lose an organ or limb, that's too bad. Your body doesn't waste time in regenerating the lost limb.
There's a lot of issues regarding the immune system also that we need to look at if scarless healing is achieved and how infection can be contained without spreading in a open wound. Regeneration takes more time to achieve, so wounds are exposed to bacteria. Nature tends to close the wound with scar tissue immediately to stop infection.

You cannot equate scarring to male pattern baldness. Baldness is not high up on researchers and physicians goals because no one dies or is completely disfigured from baldness. Scarless healing without a doubt will be achieved before anything for male pattern baldness. The argument that not finding a cure for anything is also propaganda. They just cured Hepatitis C, and are constantly researching ways to cure diseases and ailments. Look at all the plastic surgery patients surgeons will gain now that there will be no scars left behind. When you say it is natural for the body to leave behind a scar, this is true. However, PolarityTE is able to manipulate the body into NOT forming a scar. Similar to just about any other drug or treatment which is able to manipulate the body to not go to it's natural response.

Nobody dies from scarring either unless you get internal scarring which causes organ functional loss. For example, scar tissue in the heart makes you susceptible for another heart attack and scar tissue weakens your muscles.
On the skin, scars don't harm you in any way. They are a cosmetic nuisance if anything. Sometimes people get keloids but those people are few and mostly of African descent. Keloids can grow quite large and cause discomfort. As for burns scars or contracture scars, they cause immobility but with medical advancements, you can regain some movement back.
Also face transplants are becoming quite common for facial disfigurement. However, most people take anti rejection medication. That right there is enough to tell you that you have no idea how this SkinTE product can cause your body to react. Your new skin grown in some lab might be treated as foreign and your body might attack it. There is much things we still don't know about our immune systems. Even your own skin grafts can be rejected as foreign by your own body. Many burn victims cannot get skin grafts because of body rejection complications.
I can assure that up to 90% of males suffer from male pattern baldness at some point of their lives. The market for a permanent fix is pretty high. Lots of women also suffer from other types of hair loss as well. Losing hair for women can be equally painful as being disfigured.
They have few inexpensive products out there which treat hair loss but no permanent fix like Propecia. They've got nothing for scarring except super expensive lasers or skin surgery which at most gives a minor improvement.
I can assure you they know more about hair then formation of scars. I bet you baldness will be cured before your scars. All I am saying is to be cautious about Polarity or whoever publishes things on the Internet.
The only effective treatment for acne is still the very potent drug Accutane. This was discovered 40 years ago. No other equally as effective treatments have been discovered ever since. Accutane was also found by accident while attempting to treat cancer.
Why don't they focus on effectively curing acne in the first place? They prescribe you a very powerful drug and then they gauge you to fix the depressions or raised scars it leaves behind. There are many people who have serious facial disfigurement from acne. It goes to show you that if they have no permanent fixes for baldness or acne (because some people take Accutane 3 times in their lifetime to clear their skin), I bet you scarless healing is still years away.

It's probable that scarring and hair can both be cured simultaneously. Dr. Costarelis concluded that hair and skin collaberate to achieve regeneration. Tsuji has also leaned towards that. PolarityTE has shown hair regeneration from nearly scarless regenerated skin. They said it's virtually seamless so I'm guessing it's undetectable scarring which means it must be tiny.
According to them, they say your body doesn't reject it. At least in pigs they never showed anything about it being rejected. Human trials are finishing up and results should be shown soon. Either way we'll know as soon as they commercialize which they have announced and confirmed it will go into limited commercialization by the end of 2017.
I don't know why they wouldn't cure acne first either. It would probably be more easily solvable than burn wounds that can disintegrate and roast all 3 layers of the skin. But from the recent news it looks like they might've achieved success. Plus burn victims can die. If the scar shown here is really invisible to the naked eye, then people probably wouldn't care much as far as the skin market goes. Breast implants with no scars. Any surgical incisions made can be made invisible now. Any scars from accidents etc... can go unseen now. That's the hope people have with this product. Although I do wonder how they will deal with the skin biopsy.

i hope that soon we shall pass of nearly scarless at a real scar free. even if I already notice the big progress!Because I want to get rid of any memory of these scars. Forget these bad memory for good. if the scarring is invisible in the naked eye, it disturbs me not at all! But if it is a little bit visible, it would remind me these bad memory.Positive point we shall soon have the results.I am optimistic for the future !

You are so wrong when you say scars don't harm you in anyway. Maybe acne scars are just cosmetic, but burn scars which PolarityTE is aimed for are terrible. Google image some pictures and you will see there is very minimal that can be done about a constricted burn scar, the treatments they have may help movement to some degree but nothing groundbreaking. Once again male pattern baldness is not on the same research level as scarring at all. Also, technically there are things like hair transplants which can be done if you have the money right now.
People who have face transplants must take immune-suppressant drugs because the face that is now theirs comes from another human being. SkinTE uses your own autologous tissue, so 100% no chance of rejection and no need for immune-suppresant drugs. You are entitled to your opinion, but please use the facts when talking about this technology. You say be cautious of what Polarity publishes on the internet but they just received US FDA Approval and purchased a $70 million dollar facility, porcine results have been published and I have no doubt human results will be coming soon. They also have some of the brightest surgeons from the most prestigious University Hospital in the United States. But you think it's all a farce or a scam? Or even comparable to any past treatment that never even came this far?
First off, I can't say if it's a farce or not. I am just saying that they are presenting us with a great product which we have yet to see results. They posted some pics on the Internet and every one here is super extatic. There was also a paper that came out suggesting that some scientists managed to convert fibroblasts into fat cells which regenerated skin. I have no idea if both Polarity and the researchers of the paper work together.
As for baldness, scarring has nothing to do with that. The hair from the follicules stop growing because of male hormones. You will need to remove the testosterone completely from the hair follicule. This may more difficult to achieve. Hair transplants are there but in the past, they used to cut a healthy piece of skin from your scalp which resulted in a hideous scar. Now they are extracting hairs from the scalp and putting them on the bald spots.
There are many factors you need to consider such as rejection. Even if it's made from your own skin, your body can still reject it as foreign. They claim there's no rejection but proven results are yet to be seen. Also what about wound infection?
They can receive FDA grants. They are trying to develop something new. I don't see why the FDA wouldn't support them. As you said burn scars are hideous. It could help many.
All I am saying is that we can be optimistic but let's wait before we jump to conclusions.
And what happened to existing scars and scar tissue? I have some scars that are 15 years old.How canSkinTE work on these? I guess I'll put some gel on my scar and overnight my skin will be good as new with no scar in the morning.
