"Retinoids, which are vitamin A derivatives, are known to cause hypercalcemia. [7] The precise mechanism for each vitamin A compound has not been clearly defined. Retinoic acid may regulate bone cell proliferation and differentiation by suppressing alkaline phosphatase activity, osteocalcin production, and prostaglandin-induced interleukin-6 synthesis in osteoblast cells. [8] Retinoic acid also can inhibit parathyroid hormone, suggesting that increased bone resorption may account for hypercalcemia. [7]

 

The synthetic retinoid 13- cis-retinoic acid, or isotretinoin, regulates cell proliferation and differentiation and decreases N-myc expression. [9] In addition, 13- cis-retinoic acid enhances normal hematopoietic differentiation by increasing the responsiveness of myeloid and erythroid progenitor cells to myeloid colony-stimulating activity and erythropoietin, respectively. [10] It is highly protein bound, primarily to albumin, and is hepatically metabolized to 4-oxoisotretinoin, which is excreted in urine and feces. Whether 4-oxoisotretinoin is biologically active is unknown. [11]

 

Hypercalcemia has been reported in patients with neuroblastoma receiving 13- cis-retinoic acid. [12] A phase I trial of the agent in children with neuroblastoma after bone marrow transplantation examined the toxicities, pharmacokinetics, and maximum tolerated dosage. [13] Of the 51 patients between 2 and 12 years of age who received 13- cis-retinoic acid, three developed severe hypercalcemia (serum calcium levels 12.6-18.7 mg/dl) prompting discontinuation of the drug. Nine other patients had elevations in serum calcium levels that did not require drug discontinuation. The investigators concluded that hypercalcemia may be a dosage-limiting adverse effect of 13- cis-retinoic acid. Accordingly, they advised that serum calcium levels should be monitored in patients receiving high dosages of the drug.

 

A subsequent study reviewed the three patients who developed severe hypercalcemia requiring discontinuation of 13- cis-retinoic acid therapy. [12] Only one patient experienced arthralgias and myalgias; this occurred during the fifth cycle of therapy. The patient, who had been receiving 13- cis-retinoic acid 200 mg/m 2/day, also had an elevated uric acid level of 8.3 mg/dl and an increase in aspartate aminotransferase. Temporary discontinuation and a dosage reduction to 160 mg/m 2/day for subsequent cycles resolved the hypercalcemia. The second patient developed severe hypercalcemia during the second cycle of 13- cis-retinoic acid. This patient complained of tinnitus and had an elevated uric acid level of 13 mg/dl. The patient had renal insufficiency, with a reported creatinine clearance of 81 ml/minute/1.73m 2. The patient's 13- cis-retinoic acid dosage was 160 mg/m 2/day; discontinuation followed by a dosage reduction to 125 mg/m 2/day for subsequent cycles prevented further episodes of hypercalcemia. The third patient who experienced hypercalcemia (serum calcium 11.3 mg/dl) was asymptomatic and had been receiving 13- cis-retinoic acid 200 mg/m 2/day. Decreasing the dosage to 160 mg/m 2/day for future cycles resolved the hypercalcemia.

 

Our findings demonstrate that hypercalcemia may be a dosage-limiting adverse effect of 13- cis-retinoic acid in children receiving an initial dosage of 160 mg/m 2/day. To our knowledge, the only other patient reported to have severe hypercalcemia at this initial dosage had renal insufficiency. [12] Our patient had a baseline serum creatinine of 0.7 mg/dl and had no signs of renal insufficiency. A 50% dosage reduction did not prevent recurrence of severe hypercalcemia during the second cycle. We did not find any other cases in the literature of symptomatic hypercalcemia with 13- cis-retinoic acid 80 mg/m 2/day. According to the Naranjo rating scale, it is probable that 13- cis-retinoic acid caused severe hypercalcemia in our patient. [14]

 

Before each hospitalization, our patient reported a decrease in appetite and caloric intake. However, her daily dietary calcium intake was never accurately recorded. Knowledge of her calcium intake would have allowed us to exclude exogenous intake as a possible cause of her hypercalcemia. Our patient tolerated her remaining cycles of 13- cis-retinoic acid after the duration of each cycle was shortened to 7 days. Unfortunately, the efficacy of this therapy against high-risk neuroblastoma is unknown, given that the patient received a lower 13- cis-retinoic acid dosage and a shorter duration of treatment than the recommended protocol. [5]

Our findings support previous evidence that hypercalcemia is a dosage-limiting adverse effect of 13- cis-retinoic acid therapy in patients with neuroblastoma. Serum calcium levels therefore should be monitored closely in these patients. Resolution of hypercalcemia can occur with a temporary discontinuation of 13- cis-retinoic acid, followed by a 25-50% dosage reduction. Our patient required a decrease in both dosage (to 80 mg/m 2/day) and duration of each treatment cycle (to 7 days) to prevent recurrence of hypercalcemia."

Mother of God, 200mg twice per day to a child!?!?!? WTF is this study!?

Anyway some more good news. I wrote a list of people who recovered a couple of pages back. 'donteat16' was last on that list. Dubya & I weren't sure on how he recovered at the time, so just presumed it was a 'natural' recovery with time. However, Dubya messaged me last night saying how he remembered that 'donteat16' had told him a while back that he 'smoked so much weed that his parents thought that was the cause of his health problems'. So that makes another individual who recovered who also consumed a lot of ganja!

Nice. Seeing as you seem to be one of the main researchers currently on cannabis compounds. Anything of note yet in regards to eating it raw being efficacious in comparison to heating it? I've posted the video's promoting raw cannabis previously but I was wondering if there was any science behind it. I still can't seem to detach myself from evidence based stuff, anecdotal evidence doesn't always cut it for me. Plus, because you can eat it raw in far greater quantities I would get through it faster but that will make me poor very quick and I am very poor currently -_-

According to the article I'll post below, raw cannabis has lots of medical benefits but it does not activate the endocannabinoid system. There doesn't seem to be much available information on whether raw cannabis is as beneficial as heated cannabis.

For those with libido loss you can try to drink 1 (or more) glass per day/2 day of boiled parsley, 4 bunches of fresh parsley in 1L water 5 minutes boiled. It's a part of a kidney cleanse, parsley is the strongest plant to restore the adrenals, you'll see the result. It's not a permanent solution of course, it's more complicated than that and there is more to do but it's just to do the test by yourself : p

To anonyy and those of you that can answer - will the slightest exposure to an irritating food, such as wheat, dairy products, etc cancel out progress made when repairing the body? I have been transforming my eating habits in hopes that it will help my body repair itself. But from time to time, I have had something that is against my 'diet.' Do you think it reverses progress we have made or it's just like a bump in the road? I've definitely been feeling a bit better lately, for many reasons, but I don't want to deliberately compromise any improvements.

To some of you guys that have been years and years off the drug, help me out. I have had super dry skin that has stayed the same since I was on the drug 12 years ago. Most of my problems seem to relate to either lack of moisture and oil, or inflammation.

-Dry skin, irritated eyes, hair falls out with sebum plugs, keratosis pilarisis since 2003 when moved to dryer climate from coastal humid climate. In the last couple of years my hair has started to grey and I have developed wrinkles on my face far worse than my father. (I'm 28 and look f*in old and dried up) In the last year my knees crack when squat and hard to get up, muscles in my body spasm and pulse all over(like dehydration or inflammation even though drink lots water), pretty bad tinnitus in ears, and brain fog... hold on what was I saying..? j/k, frequent allergies (run in family) and headaches (since I was younger but now most are sinus headaches). I had little optimism since the beginning and couldn't bear the doom of researching so fresh after taking the drug. Now I'm at the point where I need to try something. I have been cleaning up my diet and getting ready for organ cleanses, I take borage oil and Vitamin D3 and drink smoothies everyday. (not the best diet but cleaning it up)

Heres the deal. I'm looking for some sage advice from you pseudo doctors (preferably people who have had years of experience and wisdom-not people within first few years). I am not new to this, so please save the basic stuff. I've read the whole thread since the beginning and read the science. I don't have the money to go get crap tons of tests, although I will when I do, but for now what do you guys think is my main problem? (inflamation, dehydration, toxicity, etc)

The things I have had on my list to try for awhile are:

(Baxyl) for the dehydration and dry skin

Iodine supplements(possible thyroid issue)

LDN(possible autoimmune like Lamar and movinon thought)

Has anyone else tried the baxyl or LDN? Also are there any updates about the LDN from older members?

Anyone heard anything from Lamar in awhile? (notice he stopped posting awhile ago)

Thanks guys! (Please no more diet information-I've got the picture Annoy) Think it's more than diet needed. Thx though(no disrespect)

It depend on the dosage, low dosage will only slow the progress down. But diet is just to "do no more arm" and give your body true nutrient from raw fruits & vegs. You need to help your body in elimination/detox with liver & kidney flush (intestine too) or some light method with herbs.

 

 

"Auto-immune disease" doesn't exists, it's just cells who absorbed some toxic protein (or other toxins) mainly from food and especially meat, dairy & gluten, those cells are detected by the immune system as "abnormal" or "unknown" and they are removed. Celiac disease is an inflammatory response to toxic foods, it create a leaky gut and then cause those "auto immune" reaction in the whole body.

Symptoms of Primary Hyperparathyroidism.

The general symptoms of Primary Hyperparathyroidism are listed below:

  • Low energy / fatigue.
    Generally dont feel well / Dont feel normal
    Feel older than your years.
    Lost interest in things that you used to.
    Cant concentrate, or cant keep your concentration like in the past find it difficult to read or keep concentration watching a film.
    Depression.
    Osteoporosis and Osteopenia (below normal bone density but not bad enough to be classed as Osteoporosis.)
    Bone pain usually in the bones in the legs and arms but can be other bones.
    Muscle Pain (myalgia) sore muscles, gnawing pain, burning pain, muscle stiffness if sat or stood in same place for too long.
    Joint Pain/inflammation (arthritis)
    Dehydration
    Thirst
    Frequent trips to pass urine with possible trips to toilet during the night
    Poor sleep. Broken sleep. Trouble getting to sleep.
    Tired and sleepy during the day.
    Irritable and more likely to snap at people.
    Poor short term memory (did I just put sugar in that tea?), Cant remember names for people and things easily.
    Gastric acid reflux; heartburn; GERD.
    IBS like symptoms.
    Decrease in sex drive low libido.
    Thinning hair (mainly in middle aged women on the front part of the scalp).

    Pseudo-gout (inflamed, tender painful joints caused by calcium deposits in the joints)

I understand many of these symptons are also listed with other diseases.

Yesterday I needed to go to the doctor because I decided to become a Vegetarian some months ago and now I want my blood checked out to look if my diet is ok. I mentioned the roaccutane and she said its flushed out of your system after you stop. But she was willing to check my liver levels, b12 and vit D3. She didn't want to do calcium but when any of these levels are a-normal then we check the calcium levels. Monday I get results.

I do get new pimples on my head now and my hair/hands gets oilier

I still use the CBD oil, along liver flush herbs ( which keep my intestines very calm ) and now and then some pro biotics.

P.s. I use this liver detoxer ( on top of my diet ) which has great ingredients

Interesting note (for those with sexual dysfunction)

Doneat16, the guy I mentioned the other day who I said recovered after 18 months-ish who also smoked a lot of green had the same result as me after smoking for the 1st time post-'tane;

 

"a buddy of mine hit me up to party the last night of break and i thought what the hell might as well relax one last time before i have to go to school. I got high as fuck but didnt touch any alc. i hooked up with a girl who i didnt really know or (or like for that matter) and to my amazement my dick started getting hard! i was so relieved and felt amazing

 

I stayed home and spanked it with happy tears of joy for about two weeks. these two weeks of happiness were short lived. the day after i got off grounding. it came back like a lightswitch. everything shut off like before"

 

He hadn't connected the dots (if there are any dots to connect) but he continued to smoke according to Dubya quite a lot and he eventually ended up recovering.

This is of no interest to me, but for those with inflammation/dehydration symptoms has hyaluronic acid already been tried by any of you? Nathan Carr mentions it in his 66 page report on the dangers of accutane.

Personnaly i tried hyaluronic acid long time ago (there was msm added to the supplement) and it was effective yes (skin & joint). But it's just symptomatic treatment not really something interesting.

Hey guys,

update from me: I tried the same CBD oil as Ottovisual [Edited link out]but didn't feel any different on it. After half the bottle was empty I emptied the other half in one day and still didn't feel anything. Also never felt anything from smoking/vaping (haven't tried more than a few times).

I've not found a way to consistently feel good yet, although I feel good on most mornings or if I stay inside all day. Hot showers or exercising outside make my skin dry up, my body ache and my brain feel numb. For anxiety and sexual issues I have found abstaining from masturbation and porn to be very helpful.

Your doctor is an idiot as your calcium is simply a standard test with the rest of your bloods. Meaning if she's doing a standard blood draw then it should show your calcium regardless. She ideally SHOULD be putting in a standard draw in addition to those.

    Pseudo-gout (inflamed, tender painful joints caused by calcium deposits in the joints)

    Lightheaded/dizziness/vertigo

    Poorer dental health/receding gums/increase in dental caries

    Pins & Needles (paraesthesia) or feeling of a limb "falling asleep".

I understand many of these symptons are also listed with other diseases.

Yesterday I needed to go to the doctor because I decided to become a Vegetarian some months ago and now I want my blood checked out to look if my diet is ok. I mentioned the roaccutane and she said its flushed out of your system after you stop. But she was willing to check my liver levels, b12 and vit D3. She didn't want to do calcium but when any of these levels are a-normal then we check the calcium levels. Monday I get results.

I do get new pimples on my head now and my hair/hands gets oilier

I still use the CBD oil, along liver flush herbs ( which keep my intestines very calm ) and now and then some pro biotics.

Your doctor is an idiot as your calcium is simply a standard test with the rest of your bloods. Meaning if she's doing a standard blood draw then it should show your calcium regardless. She ideally SHOULD be putting in a standard draw in addition to those.

Can you elaborate your experiences ? Did you got Hyperparathyroidism ?

And can you give us some examples of people who tested positive ?

Without jumping to conclusions a theory is maybe it compensated when we did the accutane course, and it didnt return to normal ?

For those with libido loss you can try to drink 1 (or more) glass per day/2 day of boiled parsley, 4 bunches of fresh parsley in 1L water 5 minutes boiled. It's a part of a kidney cleanse, parsley is the strongest plant to restore the adrenals, you'll see the result. It's not a permanent solution of course, it's more complicated than that and there is more to do but it's just to do the test by yourself : p

Annoy, just for the sake of clarity: you mean parsley leaves not the root, right? I believe it's "le persil" in French, right?

On April 25th, 2014, there will be a demonstration in both
the United Kingdom and the United States in protest
of the long-maintained denial of life altering and
life ending adverse effects of isotretinoin.

The U.K. demonstration will take place at Roche's
United Kingdom headquarters and will be attended by
Derek Jones, the director of the "Dying for Clear
Skin" documentary, who lost his son to this drug.

Other families who lost loved ones and several
victims of isotretinoin are also expected to attend.

The U.S. demonstration will take place at the FDA HQ
in Silver Springs, Maryland.

The main goals of the U.S. protest will be to:

- Make it be known that there are thousands, or
potentially tens of thousands left suffering severe
and persistent adverse-effects over the course of the
drugs 30 year history.

- Ask the FDA why isotretinoin is still being regularly
prescribed for moderate acne in light of the harsh
nature of the drug.

- Push for further scientific investigation into
isotretinoins full range of biological effects.
"The methods of action are unknown" is not a valid
excuse to continue denying a causal relationship to
side effects.

- Demand the FDA to require manufactures to list
persistent sexual dysfunction as a rare, but serious
possible side-effect of isotretinoin.

If you wish to attend one of these protests, and do
not want to divulge the particular side effects you
suffer from, you won't have to. Just showing up and
showing your support will help a great deal. Feel
free to bring friends or family.

This will be a good opportunity to make a difference
and finally meet others who are on the same boat.

That is awesome Dubya_B, I am glad some action is being taken about this. I would definitely go if it was closer to where I lived. Cause even though I fixed my joint problems and mental clarity, I suffer from hair loss and e.d.

Maybe this is one step closer for doctors to help find a cure for these persistent side effects.

Looks interesting. From some basic researching, it looks as though ascorbic acid (over a long period of time) and fasting and exercise increase VIP action.

Acne1776 has it spot on. It's like any other drug, it needs the right dose. Let's compare this cannabis deal with accutane. Imagine taking 0.5mg of accutane for 7 days. Do you really expect to see anything? Probably not, although saying that it wouldn't surprise me if it still f**ked some people over. Having said that I did note 7-10 days of awesomeness (close to normality sexually) after 3 days of solid vaping.

Of note was I had some banging sex last night. 100% erection, no prior stimulation just a bit of kissing etc etc. I didn't finish because it felt like I would be a one and done otherwise. But otherwise today I had another experience which I've only had since vaping which is 'finishing' but my dick stayed hard for another 5-10 minutes. This has not happened once since post-crash and before vaping. I've had it twice since I started vaping daily, consuming the after vaping bud and occasionally chewing it raw with a bit of fat. I still have no sensation though - I'm going to name this Ken Doll Syndrome from now on or KDS for short, objections? Just out of interest is anyone else suffering from complete KDS? I cannot feel my dick at all, tensing my kegel muscles to make my D twitch feels no different from puckering my butt-hole. I'm hesitant to do missionary or have my girl on top because I cannot tell if I'm hard or not, whereas with doggy-style (F yea) I have visibility. Strange thing is so far I'm only really noticing these differences with the gf, on my larry I don't feel any different down there. I guess I could test with some porn, but I do function better from abstaining for a while so I don't fap more than once or twice per week if at all nowadays. I went a solid 6 months without fapping but saw no/very minor improvement last year. So now I fap occasionally when I feel like it! And finally this time round I haven't felt the same libido as skiing, but to be honest it's too subjective to make any firm conclusions.

I'm game for going to London! Hopefully see some of you there.
